PROSPECTIVE COHORT STUDY WITH COLLECTION OF CLINICAL DATA AND SERUM OF PATIENTS WITH PROSTATE DISEASE “Etude SAKK 63/12” Sponsor : SAKK CONTACTS AUX HUG : Unité de recherche en Onco-Hématologie de la Fondation DFDL Coordinatrice de l’étude : Mme Laurence Zulianello, 079 553 25 61 Investigateur responsable: Dr Thomas Zilli, 079 553 25 63 DESIGN DE L’ETUDE Environ 2'000 patients atteints d’un cancer de la prostate seront inclus dans l’un des groupes suivants afin que du matériel (sang) et des données cliniques soient recueillies pour la biobanque (St Gall) : Groupe A : Patients programmés pour une biopsie de la prostate pour une quelconque raison - Collecte de sang avant la biopsie et en cas de re-biopsie Groupe B : Patients avec Ca Prostate traité B0 : Sous surveillance active - Collecte de sang avant le traitement curatif et en cas de re-biopsie et chaque année si pas de re-biopsie – B1 : Traité par prostatectomie radicale (RP) - Collecte de sang avant le traitement curatif et à 3 mois, 5 ans et/ou à la rechute – B2 : Traité par radiothérapie externe (EBRT) sans thérapie anti-androgénique (ADT) B3 : Traité par radiothérapie externe (EBRT) avec thérapie anti-androgénique (ADT) - Collecte de sang avant le traitement curatif et à 3 mois, 12 mois, 18 mois et/ou à la rechute - Groupe C : Patients avec une rechute biochimique après un traitement curatif avec RP : PSA en progression après RP, candidat pour une radiothérapie de rattrapage (salvage RT) - Collecte de sang avant la radiothérapie de rattrapage et à 3 mois, 12 mois, 18 mois et/ou à la rechute - Groupe D : Patients avec Ca Prostate métastatique sans traitement curatif mais avec une maladie hormono-sensible traitée avec ADT (médicale ou chirurgicale) : le patient doit être en pré-ADT au baseline puis sous ADT pour la suite. - Collecte de sang avant la castration et tous les 6 mois et/ou à la progression - Groupe E : Patients avec Ca Prostate métastatique, résistant à la castration (médicale et chirurgicale) - Collecte de sang avant la thérapie donnée suite à la résistance de la castration et tous les 6 mois, à la progression ou au changement de traitement, jusqu’au décès du patient - Etude SAKK 63/12 dngn 20.08.2015 OBJECTIF DE L’ETUDE Collecte de matériel biologique pour des projets scientifiques afin de mieux comprendre les maladies et le cancer de la prostate et de pouvoir ainsi contribuer à l’amélioration de leur pronostic et traitement. CRITERES D’ELIGIBILITE # 1 2 3 # 1 2 3 4 # 1 2 3 # 1 2 3 INCLUSION CRITERIA – GROUP A Male patient between 18 and 70 years old. Patient must give written informed consent, before registration, for storage of sera and referencing of his biopsy (-ies) in the biobank, and for clinicopathological data collection, for translational research purposes. Patient scheduled for prostate biopsy for any reason INCLUSION CRITERIA – GROUP B0 Male patient 18 years or older. Patient must give written informed consent, before registration, for storage of sera and referencing of his biopsy (-ies) in the biobank, and for clinicopathological data collection, for translational research purposes. Patient planned to be under active surveillance for localized prostate cancer All of the following criteria (according to the PRIAS study [26]) should be fulfilled: o o o o o clinical stage T1/T2 PSA ≤ 10 ng/ml PSA density < 0.2 ng/ml per milliliter biopsy Gleason score ≤ 6 one or two positive biopsy cores INCLUSION CRITERIA – GROUP B1-B3 Male patient 18 years or older. Patient must give written informed consent, before registration, for storage of sera and referencing of his biopsy (-ies) in the biobank, and for clinicopathological data collection, for translational research purposes. Patient planned to receive treatment for localized prostate cancer with either radical prostatectomy (B1), or external beam radiation therapy without (B2) or with ADT (B3) INCLUSION CRITERIA – GROUP C Male patient 18 years or older Patient must give written informed consent, before registration, for storage of sera and referencing of his biopsy (-ies) in the biobank, and for clinicopathological data collection, for translational research purposes. Patient underwent RP Patient with biochemical relapse: 4 PSA progression after RP is defined as two consecutive rises with the final PSA value > 0.1 ng/mL, or three consecutive rises (the first value must be measured earliest 4 weeks after radical prostatectomy). 5 Patient is candidate for salvage RT Etude SAKK 63/12 dngn 20.08.2015 # 1 2 3 # 1 2 3 # 1 2 3 INCLUSION CRITERIA – GROUP D Male patient 18 years or older Patient must give written informed consent, before registration, for storage of sera and referencing of his biopsy (-ies) in the biobank, and for clinicopathological data collection, for translational research purposes. Metastatic prostate cancer without curative treatment but hormone sensitive disease, planned to be treated with ADT (medical or surgical) INCLUSION CRITERIA – GROUP E Male patient 18 years or older Patient must give written informed consent, before registration, for storage of sera and referencing of his biopsy (-ies) in the biobank, and for clinicopathological data collection, for translational research purposes. Patient with mCRPC defined by: o total testosterone ≤ 50 ng/dl (≤ 1.7 nmol/l) o ongoing medical castration (if not surgically castrated) o progressive disease defined as radiological progression or PSA progression EXCLUSION CRITERIA Other concurrent active malignancy. Psychiatric disorder precluding understanding of information on study related topics and giving informed consent. Any psychological, familial, sociological or geographical condition potentially hampering compliance with the trial protocol and follow‐up. Etude SAKK 63/12 dngn 20.08.2015
