Counis Titre : Couplage du récepteur de la GnRH à la voie AMPc dans la cellule gonadotrope hypophysaire Projet de stage : : situation du sujet, objectif du stage, approches expérimentales L’équipe s’intéresse au dialogue entre la GnRH, neurohormone hypothalamique, et la cellule gonadotrope hypophysaire chez le rat. L’objectif général du stage s’inscrit dans l’étude du couplage entre le récepteur de la GnRH et une de ses voies de signalisation : la voie AMPc. Il s’agira d’étudier la régulation de plusieurs protéines, isoformes de PKC et d’adenylate cyclase, dont l’implication vient d’être démontrée au laboratoire. Les approches expérimentales viseront à quantifier l’expression de ces entités de signalisation (transcrits et protéines) dans différentes situations physiologiques (différents stades du cycle oestrien chez la femelle, traitement par la GnRH…) en travaillant à la fois avec des approches biochimiques et/ou moléculaires sur des cultures cellulaires et par des approches d’imagerie sur coupes d’hypophyse.
